Study on mechanism of morusin inhibiting glioma cells through Wnt/β-catenin signaling pathway

Abstract:

Objective To investigate the anticancer effect of moristin on glioma cells. Methods Cells proliferation was examined by CCK-8. Migration assays were performed using transwell assay. Apoptotic cells were detected using the Annexin V-FITC apoptosis detection kit. Western blotting and real time fluorescence quantitative PCR(RT-qPCR) were used to exam the change of target gene in mRNA and protein level. Results Morusin can obviously inhibited the proliferation activity of U251 cells(P<0.05), of which 100 and 1000 μg/mL had the best inhibitory effect. 100 μg/mL of morusin can obviously inhibited the migration ability of U251 cells and induce cell apoptosis(both P<0.05). Wnt/β-catenin signaling pathway was involved in anti-tumor activity induced by moristin, and morusin can down-regulate the expression of Wnt/β-catenin signaling pathway-related proteins (β-catenin and Cyclin D1(P<0.05).  Conclusion Morusin can inhibit the growth of glioma cells through Wnt/β-catenin signal pathway. Thus,morusin could be a potential anti-cancer agent for glioma.
