2020—2022年天津市布鲁菌分离鉴定及MLST分型分析

Analysis of isolation,identification and MLST typing of Brucella in Tianjin from 2020 to 2022

摘要: 目的 了解天津市布鲁菌分子流行病学特征,为今后天津市布鲁菌病(简称布病)的防控工作提供理论依据。方法 对2020—2022年天津市国家级布病监测哨点医疗机构的布病患者样本进行布鲁菌分离鉴定,应用BCSP31聚合酶链式反应、AMOS聚合酶链式反应、单因子血清凝集试验(A、M、R)对分离株进行种属及生物型鉴定,对分离株进行测序并对数据进行多位点序列分型(multi-locus sequence typing,MLST)分析,将布鲁菌公共数据和本实验研究数据一同纳入分析,用Mega XI软件进行neighbor-joining法聚类分析构建系统发育树。结果 从2020—2022年天津市国家级布病监测哨点医疗机构收集的布病患者样本中分离出12株布鲁菌,除1株为羊种1型,其余11株均为羊种3型。12株布鲁菌均为ST8型,与上海市、福建省、河北省、山西省等省市共享ST8型,与来源于贵州省的ST39型、来源于河南省和四川省的ST71型及来源于四川省的ST101型、ST118型遗传关系紧密。结论 天津市布鲁菌主要流行株为羊种3型,主要基因型为ST8型,与国内优势菌株及流行ST型一致。此为天津市首次对人源分离鉴定出的布鲁菌进行分子分型研究,为全球MLST数据库填补天津市本土数据,更好地掌握天津市布鲁菌ST型流行规律提供依据。

Abstract: Objective To understand the molecular epidemiological characteristics of brucellosis in Tianjin and provide theoretical basis for the prevention and control of brucellosis in Tianjin in the future. Methods Brucella samples from brucellosis patients in national brucellosis surveillance sentinel medical institutions in Tianjin from 2020 to 2022 were isolated and identified. BCSP31 polymerase chain reaction(BCSP31-PCR),AMOS polymerase chain reaction(AMOS-PCR) and single factor serum agglutination test(A,M,R) were used to identify the species and biotype of the isolates. The isolates were sequenced and the data were analyzed by Multi-locus sequence typing(MLST). The Brucella public data and the data of this experimental study were included in the analysis,and the phylogenetic tree was constructed with the Neighbor-joining clustering analysis using Mega XI software.Results Twelve brucellosis isolates were isolated from brucellosis patient samples collected from national brucellosis surveillance sentinel medical institutions in Tianjin from 2020 to 2022,of which one strain was brucellamelitensis biotype1,and the other 11 strains were brucellamelitensis biotype 3. The 12 strains of Brucella were all ST8,sharing ST8 with Shanghai,Fujian,Hebei,Shanxi and other provinces and cities,and were closely genetically related to ST39 from Guizhou Province,ST71 from Henan and Sichuan Province,ST101 from Sichuan Province and ST118 from Sichuan Province. Conclusion The main epidemic strain of Brucella in Tianjin is brucellamelitensis biotype 3,and the main genotype is ST8,which is consistent with the domestic dominant strain and the epidemic ST type. This is the first molecular typing study of Brucella isolated and identified from human sources in Tianjin,filling in the local data of Tianjin for the global MLST database,and better mastering the molecular epidemiology of Brucella in Tianjin.
