超高效液相色谱-串联质谱法测定面粉中8种荧光增白剂

Determination of 8 fluorescent whitening agents in flour by ultra performance liquid chromatography-tandem mass spectrometry

摘要: 目的 建立一种同时测定面粉中8种荧光增白剂(fluorescent whitening agents,FWAs)的超高效液相色谱-串联质谱法的分析方法。方法 以乙腈为提取剂,涡旋超声法提取面粉样品中的目标化合物。以2 mmol/L乙酸铵(含0.1%甲酸)缓冲液和乙腈作为流动相体系,以Phenomenex Kinetex C18柱(100 mm×2.1 mm,2.6 μm)为分析柱,采用电喷雾正离子扫描模式下的多反应监测对样品进行检测,基质外标法对8种目标化合物进行定量分析。结果 在0.10~100 μg/L范围内8种FWAs具有良好的线性关系,相关系数(r)≥0.99;FWA184的方法检出限和定量限分别为0.01、0.05 μg/kg,FWA162、FWA185、FWA367的方法检出限和定量限分别为0.03、0.10 μg/kg,FWA140、FWA199、FWA378、FWA393的方法检出限和定量限分别为0.05、0.20 μg/kg。方法的平均回收率在82.4%~113.7%之间;相对标准偏差在1.1%~6.2%之间。结论 所建立的方法简单、快速、结果准确、覆盖的FWAs种类多,适用于面粉中8种FWAs的测定。

Abstract: Objective To establish an analytical method for simultaneous determination of eight fluorescent whitening agents (FWAs) in flour using ultra performance liquid chromatography-tandem mass spectrometry. Method The target compound in flour samples were extracted with acetonitrile by vortex ultrasound method. 2 mmol/L ammonium acetate(plus 0.1% formic acid) buffer and acetonitrile were used as mobile phase for gradient elution,and the target compounds were separated by Phenomenex Kinetex C18(100 mm×2.1 mm,2.6 μm).The multiple response monitoring(MRM) was selected in the positive electrospray ionization mode to quantitatively analyze eight FWAs,and the target compounds were quantified by matrix-matched external standard method. Results A good linear relationship among 8 FWAs in the range of 0.10-100 μg/L,with a correlation coefficient(r) ≥0.99. The detection limit and quantitation limit of FWA184 were 0.01 and 0.05 μg/kg.The detection limit and quantitation limit of FWA162,FWA185 and FWA367 were 0.03 and 0.10 μg/kg.The detection limit and quantitation limit of FWA140,FWA199,FWA378 and FWA393 were 0.05 and 0.20 μg/kg.The average recoveries of the method were in the range of 82.4%-113.7%,and the relative standard deviations ranged from 1.1% to 6.2% at three spiked levels. Conclusion The established method is simple,fast,accurate and covers a wide range of FWAs,making it suitable for the determination of 8 types of FWAs in flour.
