桑辛素通过 Wnt/β-catenin 信号通路抑制胶质瘤细胞的机制研究

摘要:

目的 探讨桑辛素在胶质瘤细胞的抗癌作用。 方法 用 CCK-8 检测细胞增殖情况,利用 Traswell 试验进行细胞迁移分析,用异硫氰酸荧光素(Annexin V-FITC)凋亡检测试剂盒检测细胞凋亡情况,采用免疫印迹试验(Western blotting)和实时荧光定量 PCR(RT-qPCR)检测靶基因 mRNA 和蛋白水平的变化。 结果 桑辛素能明显抑制 U251 细胞的增殖活性(P<0.05),其中 100 和 1 000 μg/mL 桑辛素的抑制效果最佳。 100 μg/mL 的桑辛素能明显抑制 U251 细胞的迁移能力(P<0.05),并诱导细胞凋亡(P<0.05)。 Wnt/β-catenin 信号通路参与桑辛素诱导的抗肿瘤活性,桑辛素可下调 Wnt/β-catenin 信号通路相关蛋白(β-catenin 和 Cyclin D1)的表达(P<0.05)。 结论 桑辛素可通过 Wnt/β-catenin 信号途径抑制胶质瘤细胞的生长,
因此,桑辛素可能是一种潜在的胶质瘤抗癌药物。

Abstract:

Objective To investigate the anticancer effect of moristin on glioma cells. Methods Cells proliferation was examined by CCK-8. Migration assays were performed using transwell assay. Apoptotic cells were detected using the Annexin V-FITC apoptosis detection kit. Western blotting and real time fluorescence quantitative PCR(RT-qPCR) were used to exam the change of target gene in mRNA and protein level. Results Morusin can obviously inhibited the proliferation activity of U251 cells(P<0.05), of which 100 and 1000 μg/mL had the best inhibitory effect. 100 μg/mL of morusin can obviously inhibited the migration ability of U251 cells and induce cell apoptosis(both P<0.05). Wnt/β-catenin signaling pathway was involved in anti-tumor activity induced by moristin, and morusin can down-regulate the expression of Wnt/β-catenin signaling pathway-related proteins (β-catenin and Cyclin D1(P<0.05).  Conclusion Morusin can inhibit the growth of glioma cells through Wnt/β-catenin signal pathway. Thus,morusin could be a potential anti-cancer agent for glioma.
