2021—2022年新疆乙型流感病毒Victoria系血凝素抗原性及基因特性研究

Antigenicity and gene characterization of hemagglutinin genes in Victoria lineage of influenza B virus prevalent in Xinjiang from 2021-2022

摘要: 目的 了解新疆乙型流感病毒Victoria系(BV)血凝素(hemagglutinin,HA)抗原性及其基因特性。方法 从新疆15家流感监测网络实验室分离上送的BV系流感毒株中随机选取30株,以世界卫生组织推荐的BV流感疫苗株B/Washington/02/2019(2021—2022)为参考进行抗原性分析,并对上述毒株进行流感病毒全基因组测序,应用MEGA X和BioEdit软件分析HA基因进化特征。结果 与疫苗株相比,20.00%(6/30)流感毒株为类似株,80.00%(24/30)为低反应株;30株流感毒株HA基因在进化上与疫苗株亲缘关系较近,以162~164位三缺失为分支特征。与疫苗株相比,核苷酸和氨基酸同源性范围分别是93.0%~98.9%和94.0%~98.3%;30株流感毒株与疫苗株HA基因上共10个位点发生氨基酸变异,分别为D129K、R133G、E136R、E198K、A202K、K56T、T73V、Y179H、E183K、和N233D。结论 2021—2022年新疆流行的乙型流感毒株HA基因上与疫苗株亲缘关系较近,但因HA氨基酸位点发生多处变异可能导致疫苗保护效果不佳。因此,持续开展BV系流感病毒毒株抗原性分析及基因监测十分必要。

Abstract: Objective To analyze the antigenicity and genetic characteristics of the hemagglutinin(HA) gene in the Victoria lineage of the influenza B virus(BV) prevalent in Xinjiang from 2021-2022. Methods A total of 30 BV strains isolated from 15 influenza surveillance network laboratories in Xinjiang were randomly selected for antigenicity analysis using BV vaccine strain B/Washington/02/2019(2021-2022) recommended by World Health Organization(WHO) as reference,and the whole genome of the above strains was sequenced. The HA gene evolution was analyzed by MEGA X and BioEdit software. Results Compared with vaccine strains,20.00%(6/30) influenza strains were similar strains,and 80.00%(24/30) were low response strains. The HA gene of the 30 influenza strains was closely related to the vaccine strains in terms of evolution,and was characterized by three-deletion sequence 162-164. Compared with vaccine strains,the nucleotide and amino acid homologies were 93.0%-98.9% and 94.0%-98.3%,respectively. There were 10 amino acid mutations in the HA gene of 30 influenza strains and vaccine strains:D129K,R133G,E136R,E198K,A202K,K56T,T73V,Y179H,E183K,and N233D. Conclusions Most of the influenza B strains prevalent in Xinjiang in 2021-2022 are closely related to the vaccine strain on the HA gene,but the vaccine protection effect may be poor due to multiple mutations at the HA amino acid sites. Therefore,it is necessary to carry out antigenicity analysis and gene monitoring of BV strain.
